Résumé

This comprehensive review explores the role of the HER2 receptor in cancer, from its fundamental biology to its clinical implications. HER2, a transmembrane receptor tyrosine kinase (RTK), is a key player in cellular signaling, influencing cell proliferation, survival, and migration. Its activation, primarily through dimerization, triggers major signaling pathways such as PI3K/AKT/mTOR and MAPK/ERK, which are often dysregulated in cancers. Overexpression and/or amplification of HER2 are frequently observed, notably in breast and gastric cancers, making it a crucial therapeutic target.
